Ribozyme Analysis of Cis-Regulatory Elements
The core technology in all of our services is our proprietary ribozyme molecule and the required variations for specific applications. RACRE is the ONLY service and technology that is capable of discovering, isolating, measuring, and enhancing all active regulatory elements in a single reaction. Our process is the most efficient vertically integrated development and production platform for biologically derived products.
The unique mechanism of action in which the ribozyme functions in-vivo enables us to discover and enhance active regulatory elements with no prior sequence knowledge of the expressed gene or the relevant promoter regions. This unparalleled efficiency allows us to improve protein production with IP protection in weeks rather than moths or years, compared to current methods.
Our promoter enhancement service is designed to improve your existing promoter performance in the context of your production environment. This service delivers 10 novel promoters, chosen by the customer, that result from the service. This service is ideal for:
- BioTherapeutic production groups who need to increase titers and yields
- Industrial agriculture groups who need to develop specific crop protection traits
- Synthetic biology groups who need to more precisely control their pathways
- Increasing small molecule yields from biologically derived products
- Reviving abandoned drug targets due to low expression
We start this service by mutating your existing promoter and clone the resulting library into an appropriate expression system that has been modified to accept our ribozyme technology. The mutant library is delivered to your host (ie transfection, transformation, etc.) using your protocol and cultured under your conditions. After an appropriate incubation time, we recover and lyse the cell population, and sample prep for NGS sequencing.
Customers choose 10 promoters from expression levels that result from the previous step. These promoters are verified for their transcriptional activity and protein validation can either be performed by Circularis or the customer. The * denotes customer requirements (eg transformation protocol & efficiencies, plasmid maps, etc.).
Our promoter discovery service is designed to discover active regulatory elements in your host cells under your conditions. The service delivers 4 separate promoters at 10 expression levels each. The service is ideal for those researchers who are looking for:
- Constitutive promoter(s) that drive expression for a specific protein
- Additional regulatory elements to improve the expression of a protein
- Tools and methods to control multiple steps in a metabolic pathway
We first build a mutant library with genomic DNA from your host strain using an appropriate expression system that has been modified to accept our ribozyme technology. The mutant library is delivered into your host (ie transfection, transformation, etc.) using your protocol and cultured under your conditions. After an appropriate incubation time, we recover and lyse the cell population, and sample prep for NGS sequencing.
Customers choose 10 separate expression levels from each of 4 independent promoters, for a total of 40 novel promoters. These promoters are verified for their transcriptional activity and protein validation can either be performed by Circularis or the customer.
RACRE Technology Access Program
Circularis enables the success of our partners by leveraging our proprietary ribozyme technology into improved production efficiency systems. The Technology Access Program (TAP) provides customers access to the ribozyme technology for the purpose of generating novel promoter sequences where the intellectual property is owned by the customer, resulting in lower costs and dramatically lower development and financial risk.
Under the TA Program, customers gain access to the Circularis ribozyme technology and are free to generate as much promoter content as their resources will allow for a pre-determined amount of time. This flexible service-to-license business model reduces the customers development and financial risk, while providing a wide range of licensing options to fit specific business needs.
